本作品讲述了在警察“机动搜查队”，机动能力和运动神经首屈一指、但却没有刑事搜查经验也缺乏刑警常识的伊吹蓝（绫野刚 饰），观察和社交能力突出,但却不信任自己和别人的理性的刑警、志摩一未（星野源 饰），主角二人以24小时的时限内破案为目标的故事。 标题中的“MIU”是Mobile Investigative Unit（机动搜查队）的首字母，“404”是两人饰演机动搜查队员的呼叫信号。
本剧以明朝晚期的“妖书案”、“诅咒案”、“梃击案”、“红丸案”、“移宫案”等几大疑案为线索，以立储、皇权交替为关键点，讲述了明朝万历、泰昌、天启年间二十年中围绕宫廷的权力斗争，表现出君臣、父子、夫妻、同僚在权力面前交错的党争关系及体现出来的复杂情感，描绘出明朝危在旦夕的夕阳晚景。该剧着重暴露了明朝权力交替制度的弊端及吏治腐败、党派斗争、宦官专权、特务横行等一系列政治乱象，批判了黑暗的宫廷斗争，并讽刺了大臣们的愚忠。与此同时，用俯视的角度向观众展现了皇室的家庭生活，及夫妻之间、长幼之间的情感纠葛。扑朔迷离的疑案、明争暗斗的政局能吸引众多观众，而其中表现出的真实而复杂的人性，更能让人深思。
